脊髓损伤导致严重运动感觉障碍。吴周睿副教授揭示了脊髓损伤全基因组转录谱与单细胞表达谱时序性分子病理机制,明确了损伤病程节点,阐明了转录因子C/EBPβ在损伤基因网络中的核心作用。并揭示了C/EBPβ激活天冬酰胺内肽酶、调控毒性蛋白转录与传播、干扰TrkB/BDNF信号通路以诱发神经元凋亡的分子机制。研究成果在PNAS、Signal Transduction and Targeted Therapy、Molecular Psychiatry、Progress in Neurobiology、Cell Death & Disease等杂志发表SCI收录论文10篇,授权专利7项,参编专业论著2项。研究成果被Nature Reviews 系列杂志介绍及点评。主持国自然面上项目、青年项目及上海市浦江人才计划,组织并执行神经干细胞移植治疗脊髓损伤临床研究。作为骨干参与科技部国家重点研发计划、国际合作重点项目、国自然国际(地区)合作与交流项目等重大科研项目。现任国际神经修复学会青年委员。获得第十二届国际神经修复年会青年论文比赛一等奖,指导学生参与全国大学生基础医学创新研究比赛获得全国银奖、华东赛区一等奖等荣誉和奖励。
Serious motor and sensory disability results after spinal cord injury. Associate Professor Wu Zhourui explained the temporal stages of the injury process and the crucial function of the transcription factor C/EBPβ in the injury gene network. He also revealed the molecular pathological mechanism of the whole-genome transcriptional profile and single-cell expression profile of spinal cord injury, identified the molecular mechanism by which C/EBPβ activates asparagine endopeptidase, controls the transcription and spread of neurotoxic proteins, and disrupts the TrkB/BDNF signaling pathway to cause neuronal apoptosis. During these studies, he published ten papers in journals, such as PNAS, Signal Transduction and Targeted Therapy, Molecular Psychiatry, Progress in Neurobiology, Cell Death & Disease. Seven patent applications have been approved; and two academic textbooks have been published. The Nature Reviews Molecular Cell Biology, Nature Reviews Genetics, Nature Reviews Cancer and Cell Stem Cell introduced his research findings. He takes the Shanghai Pujiang Talent Program, the National Natural Science Program, and the NSFC Youth Program. He also designs and carries out the clinical trial of neural stem cell transplantation for spinal cord injury patients. As an important player, he joined great programs such as National Key Research and Development Program of China, the International (regional) cooperation and communication program (NSFC) and the State Key Program (NSFC). He serves on youth committee of the International Neurorestoration Association Research. He guided students to compete in the National Undergraduate Basic Medical Innovation Research Competition and won the National Silver Award, the first prize in the East China Division. He also won the first prize in the Youth Paper Competition of the 12th International Neurorestoration Association Research Conference (INAR).
